Peptides: Which Ones Actually Work in Skin, and How We Use Them

Peptides are the most marketed and least understood actives in skincare. The word covers everything from a two-amino-acid fragment that genuinely signals fibroblasts to make collagen, to a hydrolysed fish-skin powder in a drink, to molecules that are far too large to pass the barrier at all. Some work well; many are decoration. What a peptide is

Proteins are chains of amino acids, often hundreds long. A peptide is a short chain — by convention anywhere from two to about fifty amino acids. Collagen is a protein; a fragment of collagen three amino acids long is a peptide. The distinction matters in skin because the barrier admits small molecules and excludes large ones: a tripeptide of around 300 daltons can be pushed into the epidermis, particularly with a fatty acid attached to help it through lipid layers, while whole collagen at 300,000 daltons sits on the surface however it is marketed. Everything useful about topical peptides follows from that size rule, and so does everything misleading.

The three families that do something

Signal peptides

When collagen breaks down, the fragments themselves act as a message: fibroblasts detect them and respond by making more collagen, elastin and ground substance. Signal peptides are synthetic versions of those fragments. The best-known are a palmitoyl pentapeptide derived from a collagen sequence and a palmitoyl tripeptide and tetrapeptide pair that also calm inflammatory messengers. Applied consistently for three to six months, they produce a modest, measurable increase in dermal thickness and a softening of fine lines — real, but gentle, and slower than a retinoid. They are the peptides in our lifting infusion and in the under-eye cream we recommend, where retinoids are too harsh for the thin skin.

Carrier peptides

These bind a trace metal — almost always copper — and deliver it to enzymes that need it. The classic is a tripeptide that carries copper into the skin, where it supports the enzymes behind collagen cross-linking, wound healing and antioxidant defence. Copper peptides have the best wound-healing evidence of any topical peptide and a reasonable record for skin quality; their practical limits are cost, a blue tint, and instability alongside vitamin C and strong acids. We use them selectively in recovery routines rather than as a daily default.

Neurotransmitter-inhibiting peptides

A hexapeptide that mimics part of the protein complex nerves use to release acetylcholine can, in theory, reduce the muscle contraction behind expression lines — a very weak, topical cousin of the injected neuromodulator used in a botox treatment. In practice the effect is small, because very little reaches the muscle, and the comparison with the injection is not serious. We mention it because clients ask; we do not sell it as an alternative to the real thing.

Collagen peptides: a different conversation

Hydrolysed collagen — collagen broken into small peptides by enzymes — is the ingredient in drinks, sachets and some infusions and skin-booster formulas. Applied to the surface it cannot enter the skin and acts only as a film-forming humectant. Taken by mouth, the story is more interesting than sceptics assume: specific dipeptides and tripeptides survive digestion, appear in the bloodstream and have been shown in several controlled trials to improve skin hydration and elasticity modestly over eight to twelve weeks. The effect is real and small, and it is dose- and consistency-dependent. Injected or infused as part of a skin-booster protocol alongside hyaluronic acid, collagen peptides act as a hydrating, signalling matrix in the dermis itself, which is where they belong.

Peptides for hair

The scalp serums in our hair programmes carry a biotinoyl tripeptide — a short peptide with a biotin attached — alongside plant-derived actives that are chosen for their effect on the follicle’s growth phase. The peptide’s role is to support the anchoring proteins that hold the hair in the follicle and to prolong the growth phase; the measured effect in trials is a reduction in shedding and a modest increase in anchored hairs over a few months. It is a support to the in-clinic treatments — PRP, GFC and exosome sessions — not a substitute. Clients who use the serum consistently between sessions keep their results better than those who do not.

Peptides versus retinol: an honest comparison

Clients ask whether peptides can replace retinol. For collagen stimulation, no: a retinoid acts on gene expression in every skin cell and produces larger, better-documented changes. Peptides are gentler, work in the morning, do not sensitise to the sun and suit skin that cannot tolerate retinoids — pregnancy, rosacea, the under-eye area, a barrier in recovery. The sensible routine for most clients in their thirties and forties uses both: peptides in the morning under sunscreen, a retinoid at night.

Peptides and Jaipur conditions

  • Heat degrades peptides: a serum left in a car in May is finished. Store them indoors, in the fridge if the label allows.
  • Peptides do not sensitise to the sun, so a morning peptide serum is one of the few anti-ageing steps that is safe on a bright Jaipur day.
  • For pre-wedding plans, the lifting infusion gives a visible plumping the same day; the signal peptides at home need three months to show in the mirror — begin early.
  • Do not layer peptide serums under strong acid toners; low pH breaks the peptide bonds. Alternate nights or use the acid first and wait.
  • Hair: Jaipur’s hard water and heat make shedding worse in summer; the peptide scalp serum works better once the scalp itself is cleaned of build-up.

Who should be cautious

  • Pregnancy — signal and carrier peptides are considered safe; we skip neurotransmitter-inhibiting peptides as a precaution
  • Copper peptides with vitamin C or strong acids in the same step — they inactivate each other
  • Anyone expecting a retinoid-level result from a peptide: the effect is modest and slow
  • Very reactive skin with new products — patch test, as with any serum, though reactions are rare

What to expect

A lifting infusion shows as firmer, more hydrated skin the same day and lasts a week or two. Daily signal peptides at home show a softening of fine lines and better bounce after about twelve weeks, more at six months. Collagen peptide drinks, if you take them, improve hydration modestly over two to three months. The hair serum reduces shedding over the same horizon. None of this is dramatic on its own; all of it compounds with the in-clinic treatments it supports. Results vary from person to person.
